Fermented vegetable and fruit extract (OM-X®) stimulates murine gastrointestinal tract cells and RAW264.7 cells in vitro and regulates liver gene expression in vivo

The results of the collaborative research with associate professor Koji Wakame and his colleagues from the Department of Pharmacology at Hokkaido Pharmaceutical University School of Pharmacy have been published in the Natural Product Communications. 

 

Objective

We examined the effect of OM-X on the gastrointestinal tract cells and macrophage-like cells derived from rats, and the variation of the gene expression in the liver of the mice which were administered with OM-X in order to determine the biological effects of OM-X.

Methods

We added OM-X to the gastrointestinal tract cells derived from rats and measured the changes of production of GLP-1, which is a hormone that promotes the secretion of insulin. We also added OM-X to macrophage-like cells and measured the changes of production of nitric oxide (NO) which is an indicator of the activity of macrophages and cytokine which activates the immune system. Furthermore, we dosed the test rats with OM-X and examined their genes which showed changes in their expression level in the liver.

Results

The production of GLP-1 increased by adding OM-X to the gastrointestinal tract cells derived from rats. GLP-1 promotes the secretion of insulin which results in adjustments of the blood glucose level.

In addition, the production of NO and cytokines (IL-6TNF-α) increased by adding OM-X to macrophage-like cells. These results suggested that OM-X can activate macrophages which in turn stimulates the immune system.

Furthermore, we dosed the rats with OM-X and measured the changes of the level of gene expression in their liver. 27 out of 597 genes exhibited a change in their expression levels. The result suggested that these changes of the gene expression function as protecting cells by inhibiting inflammation and apoptosis.
